My question is, if you should fail to find a superstar card in your arsenal, but have one in your hand, can you still put it on top of Dude Love?

No. "The Mankind or Cactus Jack Superstar card" refers to the one that you found with the search. If it meant that you could use a card already in your hand, it would say something along the lines of, "You may put 1 Mankind or Cactus Jack Superstar card from your hand on top of your Dude Love Superstar card..."
